A relationship between antimicrobial peptide gene expression and capacity of a selected shrimp line to survive a Vibrio infection ArchiMer

Understanding of antimicrobial defence mechanisms of penaeid shrimp should help in the design of efficient strategies for the management and disease control in aquaculture. In this study, we have specifically analysed the expression in circulating hemocytes of antimicrobial peptides (AMPs) encoding genes, such as PEN2 and PEN3, ALF, crustin, lysozyme and a putative cysteine-rich peptide. We evidenced a relationship between the level of expression of some AMPs and the successful response of the shrimp, Litopenaeus stylirostris, to circumvent a pathogenic Vibrio penaeicida infection. Les objectifs pour l'année 1997 étaient les suivants : • Poursuivre les premières reproductions des espèces pour lesquelles nous disposons de géniteurs GO. L'objectif est toujours de limiter le stockage d'animaux importés en produisant une descendance simultanément à des témoins C. gigas qui serviront de comparaison. • Tenter des hybridations inter-spécifiques des différents taxons présents au sein du conservatoire en fonction des connaissances disponibles dans la littérature sur ce sujet (voir pour revue Gaffney et Allen, 1993). • Importer de nouvelles souches ou espèces. Élargir la gamme des espèces présentes au sein du conservatoire de souches (les capacités d'accueil de la salle de quarantaine étant limitée).

The New-Caledonian shrimp industry is based on the controlled reproduction of the shrimp Litopenaeus stylirostris, a species which was introduced in the 80s. The major difficulty to which the industry has been faced for 10 years is the occurrence of the "syndrome 93", which corresponds to mortality phases when the temperature falls down in April-May-June. This mortality is associated to the pathogenic bacteria Vibrio penaecida and is expressed at different levels which are variable from year ta year and from pond to pond. No resistance to this pathology has been developed spontaneously. Selective breeding of shrimp for viral resistance is established for species like L. vannamei and L. stylirostris. However, selection for bacterial infection resistance is not documented. In New Caledonia, "Syndrome 93" is a vibriosis that affects cultured shrimps during the cool season with survival from 15 to 30%. Current practises for breeders rearing at low densities do not improve the resistance of the offsprings mainly because selection pressures for "Syndrome 93" are low. Two oyster species are currently present along the French coasts : the indigenous European flat oyster (Ostrea edulis), and the Pacific cupped oyster (Crassostrea gigas), that has been introduced from Japan since the beginning of the 70ies. The flat oyster successively suffered from two protozoan diseases during the 60ies and its production decreased from 20 000 tons/year by that time to 1 500 tons/year nowadays. Consequently, the oyster production is principally (99%) based upon the Pacific oyster species with approximately 150 000 tons/year among which 90% are grown from the natural spat. Pendant l'été 1995, des mortalités d'huîtres creuses ont été observées sur une grande partie du littoral français et notamment en Bretagne. Pour cette région, un ensemble d'informations concernant ces mortalités a été recueilli au près d'organismes en contact direct avec les professionnels : IFREMER, les Affaires Maritimes et la Section Régionale Conchylicole de Bretagne sud. L'objectif de ce rapport est, à partir de quatre sources d'informations, de chiffrer ces mortalités dans les différents sites bretons et d'étudier les relations éventuelles avec différents paramètres tels que l'origine des huîtres, la technique d'élevage, le site d'élevage, etc... As of 1985, Ifremer set up a selection programme for resistance to the protozoan Bonamia ostreae in European flat oysters (Ostrea edulis). Two selection lines, L85 and L89, from the third and second generation, respectively, were produced with 17 and 21 different families. The experimental design also included a mixed line (L85 x L89) of seven families and a control line of five families from Quiberon broodstock maintained without selection pressure.

Le programme de sélection de l'huître plate a été initié en 1985 par des pathologistes de la Tremblade. La sélection ne portait que sur le seul critère de tolérance des huîtres plates au parasite Bonamia ostreae et correspondait plus à un outil pour étudier divers aspects de la parasitose qu'un un programme d'amélioration génétique visant la relance de la culture de l'huître plate. Dans le cadre du réseau REGEMO, ce programme a ensuite été repris par les généticiens de l'Ifremer : 1) rappel historique et définition d'un vocabulaire commun à l'ensemble des membres du réseau REGEMO. 2) Inventaire et bilan des opération menées ou en cours ( sur la dernière génération de sélection produite ne 1995, sur le matériel issu de cette génération et produit en 1997....

Genetic structure at different spatial scales in the pearl oyster (Pinctada margaritifera cumingii) in French Polynesian lagoons: beware of sampling strategy and genetic patchiness ArchiMer

In order to study further the genetic structure of the pearl oyster Pinctada margaritifera in French Polynesia with a special consideration for the sampling scale, we analyzed or re-analyzed sets of data based on nuclear DNA markers obtained at different spatial scales. At a large scale (several 1,000 km), the remote Marquesas Islands were confirmed to be significantly differentiated from Tuamotu-Gambier and Society archipelagos, with a marked difference however for the two main islands that are different from each other. The tropical seabass Lates calcarifer was introduced in Tahiti in 1984 and then was domesticated without other introduction. Three successive generations have been obtained in captivity. Tissue samples of the 38 animaIs which represent the total Tahitian broodstock were preserved in alcohol to be genotyped with four microsatellite markers (Yue et al., 2001). Three of four described markers were successfully transferred to the laboratory of genetics of Tahiti (marker LcaM01, LcaM02, LcaM03), as the revelation technology available locally did not allow the use of LcaM04 in routine. The Aquaculture and Pathological Genetics (GAP) laboratory at La Tremblade has been monitoring three populations of European flat oyster since 1985 or 1989 under selection for higher survival rates in the presence of the parasite Bonamia ostrea. One selection cycle (1985 population) or two (1989 populations) were carried out by mass selection by placing the oysters for several years in a highly infested zone, and then harvesting spawns from survivors.

Peneid shrimp are commonly reared in clear-water aquaria or tanks for short-term studies of 4-6 weeks during controlled experiments, such as nutrition studies to estimate dietary nutrient requirements. Recently, in line with the genetic program at Centre Oceanologique du Pacifique (COP), experimental clear-water facilities were tested for the first time over a longer rearing period. Environmental conditions used to maintain animal growth in clear-water system using a well-balanced diet were defined. The European flat oyster Ostrea edulis has been considered as the traditional French oyster for a very long time and has now nearly disappeared from our farms, as a consequence of the two successive epizootic of the Marteilia refringens parasite and Bonamia ostreae. The actual quasi monoculture of Crassostrea gigas has made necessary the search for a diversification in oyster culture, including the search for a new start of the indigenous flat oyster culture. The Ifremer has been conducting parallel works since 1985 in order to better understand the biology of the parasite as well as to try and select different lines of flat oyster resistant to Bonamia ostreae.

The network IFREMER/REMORA has watched every year, since 1993, from February to December, mortality, growth and quality criteria of two oyster class-size ("juveniles" of the first-year c1ass and "adults" of the second year c1ass), distributed among various stations of the French oyster areas. This has provided a quite standard and simple annual assessment of the rearing results, generally tallied with professional observations. From year to year, data series have been obtained, from which mean values (references), chronological trends or spatial differences can be analysed. L'aquaculture en Martinique commence dans les années 1970 avec le transfert technologique de l'élevage de la chevrette : Macrobrachium rosenbergii. Au début des années 1980, le CNEXO en collaboration avec une association régionale (ADAM) travaille aux Antilles à la sélection d'espèces tropicales de poissons. Cette étude s'inserre alors dans un programme international de sélection d'espèces tropicales, coordonné par le Centre Océanologique du Pacifique.
